I know how a viscous coupling works but would a full tread depth tyre to min depth of 2% difference in circumference really knacker a diff on a motorway journey?

I have swapped my wheels around but just for even wear on the tyres. I'm sure there are many out there who wouldn't even do that and I've personally not heard of any centre diffs failing.

Just to throw something else in the mix to keep you lot arguing lol... What about running spacers on the rear only? all wheels exactly the same size,width,tyres and brand etc... only difference being spacers on the rear?

Spacers will not cause an issue with the diff. You will here people say that the extra offset caused by running spacers will put additional strain on your wheel bearings but no more than running high offset wheels
