hi mate,
I take it you're talking about the SAM mission?
I did it by stealing a fairly sturdy / fast car, taxi for example. Then scream over to the dock bit to get the boat. Drive down the walk way (cos you need a quick getaway when you get back to shore)
Grab the rocket launcher and jump in the boat, drive to the bouy and wait for the plane.
you have to be quick to shoot it down. You want all the packages to fall in the water. Then pick em all up (I think you get a star for each).
A helicopter will no doubt get to you in no time, resist the temptation to shoot it down.
Get back to the car asap and then jump in and leg it back to the bodyshop to remove your stars then you can simply drive back to the correct location at your leisure.
Alternatively, one of the guys at work said that you can drive to the airport, kill all the waiting Columbians and then allow the plane to land. Get the packages and then leg it to the nearest bodyshop. Again, simply drive back to the location on the map.

the only time you can get more than one ho in the car is when you're doing the Luigi mission "Fuzz Ball". If you get a mini van type car you can get 4 or 5 in at once
The strangest thing I've managed is driving a stretch and picking up a ho in the back and they still manage to get it on.

I spent a while trying to get the best way out of that one, one method I used was to get the linerunner(truck) from the cafe opposite and remember where all the stars are, then once you get out of the boat you have to have a fairly sturdy vehicle to smash up the FBI cars, gotta be quick though...
I think I also got the fast car over by Asukas place and out run them whilst getting the stars....

that mission... D'OH!
soz... that's not too hard I remember.
it's pretty much the same as SAM... the secret is to go to the paint shop when you get back to shore. It's right next to Asuka's place anyway.
