ok, needin some help;

Firstly, I can pick up a healthpack, but it doesn't affect the bar on the right hand side, am I being thick? Do I need to activate it in some way!?!

Also, mission help; second or third mission, the one where you're in the defence ministry building. I've got the computer info and am trying to extract - but where too???? The rooftop is deserted, and I've tried climbing down the liftshaft a couple of times but to no avail... arrgth!

you need to go into your inventory (where you change guns etc) Cursor over to the left and you'll see your health packs. just select it and you'll be rewarded with a "ahh" and the bar going up

On the rooftop, you need to approach the left hand edge of the building and there is a small chiney that will allow you to rappel off. (Well, assuming you are talking about the same bit).

If you are talking about the roof where you have to 'imbolise' a guard then the way out is down that lift shaft. You just have to keep dropping down ledge by ledge to the bottom.
