Here's what I'd do:
Plug in your iPod and launch iTunes. Configure the settings for your iPod to ensure it's set to manually manage music. Unplug your iPod and plug in the wifes iPod and do the same thing. This will stop iTunes from automatically syncing the iPod(s) against the music library.
If the music on your iPod exists in the iTunes library then you can simply copy it to her iPod. If it's not on the library and just on your iPod then iTunes isn't going to help as you cannot copy from iPod to iTunes, you'll need third party software as mentioned by bioforger.

Do you each have your own login?
If so, you can have a copy of itunes on each but both reference the same shared music folder - you'll then have to go through each track in the library and uncheck / check the boxes next to each track to update on each login.
A pain I know buts its 1 way...
