I've just found my Settlers 3 cd's knocking around in the cupboard and thought it'd be fun to have a quick game, however, after installing it, Vista refuses to load it. I'm guessing this is as it's such an old game and designed for a DOS based O/S.
Is there anyway to make this game work on my Vista driven machine?

It is doable, you will need either an emulator or run a virtual pc inside Vista.
I would do the latter, download Virtual PC here..
Windows Virtual PC: Home Page
and Dos here..
Download details: MS-DOS 6.2 Step-up Files
Both free.
